Physiologists from Russia and Germany were the first to identify the key role of the recently described family of potassium channels TASK-1 in the regulation of arterial tone and blood pressure in newborns. The discovery will help develop strategies for the treatment of cardiovascular diseases in children. The work was published in the British Journal of Pharmacology.

Researchers have demonstrated for the first time a high representation and pronounced functional activity of TASK-1 potassium channels in the arteries of newborn rat pups, where they contribute to the maintenance of blood pressure at a low level. These channels belong to the family of potassium channels with two pore-forming loops and were relatively recently discovered in the vascular bed.

In a newborn, the level of blood pressure is almost two times lower than that of an adult, in many respects it is determined by the state of the arteries of the resistive type. These arteries, despite their small diameter, are capable of changing the size of the internal lumen in a wide range. In a newborn organism, the resistive arteries must be in a more relaxed state in order to reduce the stress on the still immature heart and to protect the vascular walls from excessive stretching. Therefore, the mechanisms of regulation of the tone of resistive arteries in the newborn organism have a number of distinctive features.

“The experiments were carried out on laboratory animals - rats. We found a higher level of TASK-1 channels in the arteries of newborn rats than in adult rats, says Anastasia Shvetsova, co-author of the work, a researcher at the Department of Human and Animal Physiology at Moscow State University. - Blockade of the TASK-1 channels led to the development of contraction and pronounced depolarization (a decrease in the potential difference) of the smooth muscle membrane of isolated segments of the arteries of rats, but not of adult rats. At the systemic level, we found that intravenous administration of a TASK-1 channel blocker caused an increase in blood pressure only in rat pups. The body of data obtained from both molecular biological studies and functional measurements indicates an important physiological role of TASK-1 channels in maintaining a more relaxed state of smooth muscle of arteries in the newborn organism. This fact must be taken into account when developing strategies for the treatment of cardiovascular diseases in children, the frequency of which has been steadily increasing in recent years.
